A review of the complexity of biology of lymphatic filarial parasites2009 •
There are about five more common, including Wuchereria bancrofti and Brugia malayi, and four less common filarial parasites infecting human. Genetic analysis of W. bancrofti populations in India showed that two strains of the species are prevalent in the country. The adult filarial parasites are tissue specific in the human host and their embryonic stage, called microfilariae (mf), are found in the blood or skin of the host, depending upon the species of the parasite. Three genetically determined physiological races exist in W. bancrofti and B. malayi, based on the microfilarial periodicity. They are the nocturnally periodic, nocturnally subperiodic and diurnally subperiodic forms. The susceptibility of a mosquito species to filarial infection depends on various factors, which could be genetic, physiological or physical. Size and charge antigens of Dirofilaria immitis adult worm for IgG-ELISA diagnosis of bancroftian filariasis2010 •
We used Dirofilaria immitis adult worm antigens to develop an IgG-ELISA, then used this to evaluate 30 serum samples of patients with proven Wuchereria bancrofti infection, 131 samples of patients with other parasitic diseases and 30 serum samples of healthy controls. The D. immitis antigen was prepared using two methods: Sephacryl S-200 chromatography and iso-electric focusing with a Rotofor cell. This was done to determine the best method for diagnosing W. bancrofti filariasis. Before fractionation, crude male D. immitis antigen yielded 100% sensitivity and 60.8% specificity, and crude female antigen yielded 80% sensitivity and 52.8% specificity, respectively, to detect W. bancrofti infection. After gel filtration chromatography, the male D. immitis antigen, called MP1, yielded 100% sensitivity and 95% specificity, and female D. immitis antigen, called FmP1, gave 100% sensitivity and 59.6% specificity, to detect W. bancrofti infection. BACKGROUND INFORMATION Schistosomiasis, also referred to as "bilharziasis" after Theodor Bilharz who first identified the parasite first in 1852 is caused by infection with parasitic blood flukes known as schistosomes. This parasite is thought to have been brought to the western hemisphere in the times of the African slave trade. This is when a host of vulnerable may have been introduced, most likely in the casks of drinking water that were brought with the slaves (Bogtish, et al., 2005). This blood fluke has 2 different hosts, the first one being most fresh water snails and the second being human, mice and hamsters. MORPHOLOGY These blood flukes have five different developmental stages. They are eggs, miracidia, sporocyst, cercariae and adult worms. Eggs are round to oval in shape, operculate and contain a developing embryonic larva (miracidium). Differences in egg morphology can be used to distinguish between Schistosoma species: S. mansoni producing oval eggs (115-175 x 45-7µm) with a sharp lateral spine. Miracidia are elliptical free-swimming larval stages (~200µm long) covered with cilia. Sporocysts appear as pleomorphic sac-like bodies which contain developing cercariae. Mature cercariae are elongate free-swimming larval stages (400-600µm long) consisting of a tapering head (with prominent penetration glands) and a forked tail (furcocercous). Adult flukes are elongate tubular worms (10-20mm long), with rudimentary oral and ventral suckers. Males are shorter and stouter than females, and they have a longitudinal cleft (gynecophoral canal or schist) in which the longer slender female lies folded.
